Analyzing of measured solar spectrum

This diploma thesis deals with the definition of solar lights for solar purposes and solar simulators. These concepts are presented and described in detail. The theoretical part is described below chapter explaining the possibilities of using solar energy phototherapy. The next section follows the issue of standards and their podnorem AM, describes in detail the direct and diffuse light component. In connection continues with IEC 60904, IEC 60904-3 and IEC 60904-9, differences in purpose and use. The thesis describes the problems of the methods of analysis of the solar spectrum as defined in IEC 60904-9. In the penultimate chapter we discuss all light sources, their properties and uses. They navyzují selected light sources used in this work, as well as descriptions and color filters designed for measurement and subsequent evaluation in the practical part. The last chapter analyzes used measuring instruments, their advantages and disadvantages. In the practical part, we describe a method for measuring real-world light spectra, determining compliance with these spectra spectra AM 1.5 and the necessity to generate data from IEC 60904-3. Another part allows measured data to see in an understandable form and must include an assessment and assignment of the data set into classes according to AM 1.5.
Design of Biolamp

This thesis describes relationship between human body and light. It is focused on perception of light with human eye, skin and their reaction to incident light and its intensity. Then it contains short description of Icterus Neonatorum (yellow-gum), Constitutional Dermatitis, Lepriasis and their treatment. It is followed by description of various sources of light, their use in medicine and reaction of body to them. This work includes measurements and evaluation of five sources of light.
Analysis of bilirubin's relevant photoproducts and their biological significance in the issue of neonatal jaundice

Neonatal jaundice occurs in almost 60% of full-term and 80% of premature babies, where a slightly increased concentration of bilirubin protects against oxidative stress just after birth. When a specific bilirubin level is exceeded in serum (usually above 340 µmol·l-1 ), bilirubin-could induce kernicterus. These negative states are prevented by blue-green light phototherapy (420-510 nm), which converts bilirubin into more polar photoproducts that are more easily excreted via bile and/or urine. Published data have shown that newborns with indicated phototherapy may develop clinical problems later in life (higher incidence of e.g. asthma, allergies, type 1 diabetes was observed at a later age). A possible reason for the occurrence of these diseases is the specific biological activity of photoproducts. This study aims to purify an unknown photoproduct (band P), formed after 8 hours of irradiation of a bilirubin solution with blue light, and to establish a quantitative analytical method for its measurement in relevant matrices. This product was isolated and separated by a thin layer and subsequent column flash chromatography. In the next part, an LC-MS/MS method was developed for quantification of band P. Study of bilirubin and its oxidation products

Phototherapy (PT) with blue-green light (420-490 nm) is the standard treatment for severe neonatal jaundice to prevent infants from toxic bilirubin (BR). Upon blue-green light exposure, BR is converted to more polar photoisomer (PI) lumirubin (LR) and the other oxidation products (mono-, di-, tripyrrols) which can be more easily disposed of the body via urine and/or bile. Although generally considered to be safe, PT is accompanied by an increased risk of various pathophysiological conditions (inflammatory processes, allergies, diabetes, and some types of cancer), in extremely low-birth-weight newborns. Thus, to account for these consequences, our study aimed to understand the mechanism of BR secretion in different tissues and cell lines and investigate the bioactive properties of BR and its main photooxidation product LR. At first, we focused on the detection of BR in the bile and feces of hyperbilirubinemic Gunn rats. Simultaneously, we tested the antioxidant and pro-oxidant effects of unconjugated BR in human hepatoblastoma (HepG2), proximal tubular (HK2), neuroblastoma (SH-SY5Y), and murine endothelial (H5V) cells by exposing them to progressively increasing concentrations of BR. Present work has been focused on the importance of the products of the heme catabolic pathway, in particular under conditions of unconjugated hyperbilirubinemias (neonatal jaundice and Crigler-Najjar syndrome (CNS)). The second part of the project was focused on the improvement of some pharmacological approaches used in the treatment of these diseases, as well as on studies of bilirubin products that are formed during the treatment by phototherapy (PT). Neonatal jaundice is one of the most common complications in neonates. Currently, there is no efficient pharmacotherapy and the treatment with blue light is used as a gold standard for severe neonatal jaundice. However, the absolute safety of PT has still not been confirmed. In this context, it is important to note that some neonatologists start the PT before serum bilirubin levels reach the recommended values and that patients with CNS type I (CNSI) are forced to be on life-long PT (unless undergoing liver transplantation). The focus of the present project was to study biological effects of bilirubin photoisomers (PI) in an in vitro model of the human neuroblastoma SH-SY5Y cells that are used for studies of the neuronal metabolism.
